脑线粒体形态和呼吸链参与D-半乳糖诱导脑衰老的研究

Brain mitochondrial dysfunction in D-galactose-induced brain aging

  • 摘要: 目的:观察脑线粒体形态和呼吸链是否参与了 D-半乳糖诱导的脑衰老过程。方法采用 D-半乳糖制备动物衰老模型,生化分光光度法检测小鼠脑线粒体呼吸链复合物的活性,透射电镜下观察海马结构内线粒体超微结构的改变。结果模型组较对照组脑线粒体呼吸链复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性下降。透射电镜下,模型组线粒体超微结构有明显病理改变。结论脑线粒体形态异常和呼吸链复合物活性下降参与了 D-半乳糖诱导脑衰老的过程。

     

    Abstract: Objective To observe the involvement of mitochondrial dysfunction in D-gal-in-duced aging model.Methods Respiratory chain activities (complex Ⅰ,Ⅱ,Ⅲ and Ⅳ)in brain mitochondria and ultrastructural impairments of hippocampal mitochondria were examined in the mice, which received 6 weeks D-galactose injection.Results Compared to the control group,D-galactose-injected mice showed decreased levels of brain mitochondrial respiratory chain and ultrastructural path-ological changes of hippocampal mitochondria.Conclusion Brain mitochondrial degeneration may be involved in D-galactose -induced brain aging.
