五子衍宗方对豚鼠形觉剥夺性近视眼巩膜形态学变化的影响

Effect of Wuzi Yanzong Recipe on change of scleral morphology in guinea pig with form-deprivation myopia

      目的  观察豚鼠屈光状态、眼轴及巩膜形态学变化,了解五子衍宗方对形觉剥夺性近视眼的影响。
      方法  选择2周龄健康豚鼠100只,通过眼罩遮盖右眼42 d制备形觉剥夺性近视模型,测量并记录实验豚鼠双眼屈光度和眼轴,将造模成功的豚鼠随机分为对照组和治疗组,再次测量记录双眼屈光度及眼轴。治疗组使用五子衍宗方水煎液(生物给药量为16 g/kg)灌胃, 1次/d, 连续30 d, 对照组不进行干预。采用电镜检查2组模型眼巩膜组织的超微结构并比较差异。
      结果  实验豚鼠经遮盖42 d后,右眼成功诱导出形觉剥夺性近视眼模型,与自身对照眼相比,眼轴与屈光度均有显著增加(P < 0.05)。电镜结果显示, 2组豚鼠近视眼模型后部巩膜形态均有不同程度改变,相较于治疗组模型眼,对照组模型眼的结构改变更为明显。
      结论  五子衍宗方可能具有延缓形觉剥夺性近视眼豚鼠巩膜组织形态改变进而延缓近视进程的作用。

     

    Abstract:
      Objective  To observe changes of refractive status, axis oculi and sclera morphology in guinea pigs and to explore the effect of Wuzi Yanzong Recipe on changes of scleral morphology in guinea pig with form-deprivation myopia.
      Methods  A total of 100 healthy guinea pigs with 2 weeks of age were selected, and deprivation myopia models were prepared by eye mask covering for 42 days. Their binocular diopter and axis oculi were recorded. The successful models were randomly divided into control group and treatment group, and their binocular diopter and axis oculi were recorded again. The treatment group applied intragastric administration of Wuzi Yanzong Recipe at a dose of 16 g/kg, once daily for 30 consecutive days. No intervention was performed in the control group. The sclera ultrastructure of the treatment group and the control group were examined and compared by transmission electron microscopy.
      Results  The right eyes of guinea pigs were successfully induced for form-deprivation myopia model after 42 d covering. Compared with the self-controlled eyes, the eye axis and diopter were significantly increased (P < 0.05). The electron microscope results showed that posterior scleral morphology in the treatment group showed some pathological changes, but showed anobvious changes than those in the control group.
      Conclusion  Wuzi Yanzong Recipe can delay progression of myopia by postponing the changes of scleral tissue morphology.
