三氧化二砷诱导急性淋巴细胞性白血病细胞系凋亡

ARSENIC TRIOXIDE CAN INDUCE APOPT0SIS OF ACUTE LYMPHOCYTIC LEUKEMIA CELL LINE

  • 摘要: 目的:了解三氧化二砷(As2O3)对急性淋巴细胞性白血病细胞系的可能作用。方法:以急性淋巴细胞性白血病细胞系MOLT-4为体外模型,在观察As2O3对细胞生长、活力及形态学影响的基础上,应用流式细胞仪检测细胞DNA含量分布,以评判细胞凋亡与否。结果:As2O3抑制MOLT-4细胞的生长呈浓度依赖性,1.0μmol/L,的As2O3诱导MOLT-4细胞凋亡。结论:As2O3能有效地诱导急性淋巴细胞性白血病细胞系调亡.并可能为As2O3在临床上治疗此类患者提供理论依据。

     

    Abstract: Aim: To understand the possible effect of arsenic trioxide (As2O3) on acute lpophocyt-ic leukemia cell line. Methods: Acute lymphocytic leukemia cell line MOLT-4 was used as in vitro model.On The effects of AS2O3 on cell growth, viability and m0rphology, apoptosis was assessed by means of rnor-phology, distribution of DNA contents on flow cytometry. ResultS: l. 0mol/L of AnO3 induced MOLT-4 cell apoptosis in the time-dependent manner. Conclusions: It may suggest that An O3 can induceapoptosis of acute lyrnphocytic leukemia cell line and may privode a theoretical base in clinic.
