Expression of phosphatase and tension homology deleted on chromosome ten and phospho-protein kinase B protein in gastrointestinal stromal tumor and their effects on the prognosis of patients

  •   Objective  To investigate the expression levels of phosphatase and tensin homology deleted on chromosome ten (PTEN) and phospho-protein kinase B (p-PKB, also called p-Akt) in gastrointestinal stromal tumor and and their effects on prognosis of patients.
      Methods  A total of 116 patients with gastrointestinal stromal tumor undergoing surgical treatment were selected as study group, and the corresponding normal adjacent gastrointestinal tissues were selected as control group. The expression levels of PTEN and p-Akt in the two groups were determined by immunohistochemistry; the relationships between the expression levels of PTEN, p-Akt and the clinicopathological features of gastrointestinal stromal tumor were analyzed; the relationships between the expression levels of PTEN, p-Akt and relapse-free survival of patients with gastrointestinal stromal tumor were analyzed; the prognostic factors of gastrointestinal stromal tumor were investigated.
      Results  The positive expression rate of PTEN in the study group was significantly lower, while the positive expression rate of p-Akt in the control group was significantly higher than that in the control group (P < 0.05). The low expression of PTEN and high expression of p-Akt were related to the mitosis, risk grade and invasion depth of gastrointestinal stromal tumor (P < 0.05). The mean recurrence-free survival time of patients with PTEN positive gastrointestinal stromal tumor was significantly longer than that in patients with PTEN negative gastrointestinal stromal tumor (P < 0.05); the mean recurrence-free survival time of patients with p-Akt negative gastrointestinal stromal tumor was significantly longer than that of patients with p-Akt positive gastrointestinal stromal tumor (P < 0.05). Pathological mitosis, depth of infiltration, negative expression of PTEN and positive expression of p-Akt were the influencing factors for prognosis in patients with gastrointestinal stromal tumor (P < 0.05).
      Conclusion  The expression of PTEN and p-Akt are closely related to the prognosis of gastrointestinal stromal tumor, which can provide the reference for predicting the prognosis.
